Plasmids containing the same origin of replication are useful tools to perform biotechnological studies in Pseudomonas putida U and in E. coli DH10B

چکیده
Plasmids containing the same origin of replication (pBBR1MCS-2 Km and pBBR1MCS-3 Tc) have been used to express simultaneous and independently different proteins in P. putida U and in E. coli. Thus, when P. putida was transformed with different genetic constructions made in the same plasmid (pBBR1MCS-3 Tc), or with plasmids containing the same replication origin but with different antibiotic resistant genes (Km and Tc), they co-existed inside the same microbe. Furthermore, when E. coli DH10B was transformed with the plasmids recovered from the recombinant P. putida U, we noticed that all the bacteria isolated from single colonies are resistant to Km and Tc, suggesting that these plasmids were also present in E. coli. This observation facilitates the genetic manipulation of these strains (i.e. avoiding the use of different plasmids in double or multiple complementation experiments), and could be an interesting tool to approach many metabolic and biotechnological studies.

متن کاملVaricella Zoster Virus (VZV) Origin-Dependent Plasmid Replication in the Presence of the Four Overlapping Cosmids Comprising the Complete Genome of VZV
The Varicella-Zoster Virus (VZV) genome contains both cis-acting and trans-acting elements, which are important in viral DNA replication. The cis-acting elements consist of two copies of oriS, and the trans-acting elements are those genes whose products are required for virus DNA replication. It has been shown that each of the seven genes required for ori-dependent DNA synthesis of Herpes Simpl...
متن کاملPseudomonas chromosomal replication origins: a bacterial class distinct from Escherichia coli-type origins.
The bacterial origins of DNA replication have been isolated from Pseudomonas aeruginosa and Pseudomonas putida. These origins comprise a second class of bacterial origins distinct from enteric-type origins: both origins function in both Pseudomonas species, and neither functions in Escherichia coli; enteric origins do not function in either pseudomonad. Both cloned sequences hybridize to chromo...
متن کاملRoles of long and short replication initiation proteins in the fate of IncP-1 plasmids.
Broad-host-range IncP-1 plasmids generally encode two replication initiation proteins, TrfA1 and TrfA2. TrfA2 is produced from an internal translational start site within trfA1. While TrfA1 was previously shown to be essential for replication in Pseudomonas aeruginosa, its role in other bacteria within its broad host range has not been established. To address the role of TrfA1 and TrfA2 in othe...
متن کاملEffects of the presence of ColE1 plasmid DNA in Escherichia coli on the host cell metabolism
BACKGROUND Although understanding of physiological interactions between plasmid DNA and its host is important for vector design and host optimization in many biotechnological applications, to our knowledge, global studies on plasmid-host interactions have not been performed to date even for well-characterized plasmids. RESULTS Escherichia coli cells, either devoid of plasmid DNA or bearing pl...
